  • 目的 探讨小鼠心肌梗死进程中巨噬细胞亚群的动态变化与CD59、P2X7表达的动态变化。方法 将成年雄性C57BL/6J小鼠36只随机分为对照组(不做任何处理,6只)和心肌梗死组(30只)。通过冠状动脉左前降支结扎建立小鼠急性心肌梗死模型,心肌梗死组30只小鼠又随机分为A、B、C、D、E组,分别在心肌梗死后1、3、5、7、14 d取材。收取小鼠的外周血和心脏组织,采用流式细胞术检测巨噬细胞亚群的动态变化与CD59和P2X7的表达情况。结果 心肌梗死后1、3 d外周血中单核细胞数量均高于对照组[(9.5±1.7)、(10.3±3.7)×104/ml比(3.2±3.1)×104/ml](均P<0.05)。心肌梗死后1、3、5 d心脏组织白细胞计数、巨噬细胞计数均高于对照组(均P<0.05)。心脏组织中Ly6Chigh巨噬细胞计数在心肌梗死后3 d达到高峰,而Ly6Clow亚群在心肌梗死后5 d达到高峰,均高于对照组(均P<0.05)。心肌梗死后1 d P2X7-CD59-巨噬细胞占Ly6Chigh巨噬细胞百分比高于对照组,而P2X7+CD59+巨噬细胞占比低于对照组(均P<0.05)。心肌梗死后1、3、5 d P2X7+CD59+巨噬细胞占Ly6Clow巨噬细胞百分比均低于对照组,而P2X7-CD59+巨噬细胞占比均高于对照组(均P<0.05)。结论 在小鼠急性心肌梗死的不同阶段,心脏组织巨噬细胞的亚群呈现动态变化,同时伴随CD59和P2X7表达的动态变化。

  • Objective To investigate the dynamic changes of macrophage subpopulations and the expressions of CD59 and P2X7 during the process of myocardial infarction in mice. Methods A total of 36 adult male C57BL/6J mice were randomly divided into control group (6 mice without any treatment) and myocardial infarction group (30 mice). A mouse model of acute myocardial infarction was established by ligating the left anterior descending coronary artery, and 30 mice in the myocardial infarction group were randomly divided into groups A, B, C, D, and E, and the samples were collected at 1, 3, 5, 7, and 14 d after myocardial infarction, respectively. The peripheral blood and heart tissues of mice were collected. The dynamic changes of macrophage subpopulations and the expressions of CD59 and P2X7 were detected by flow cytometry. Results The numbers of monocytes in peripheral blood on the 1 and 3 d after myocardial infarction were higher than those in the control group [(9.5±1.7), (10.3±3.7)×104/ml vs (3.2±3.1)×104/ml](all P<0.05). The white blood cell counts and macrophage counts of heart tissue at 1, 3 and 5 d after myocardial infarction were higher than those of the control group (all P<0.05). The Ly6Chigh macrophage count in cardiac tissue peaked at 3 d after myocardial infarction, while the Ly6Clow subset reached a peak at 5 d after myocardial infarction, both of which were higher than those in the control group (all P<0.05). The percentage of P2X7-CD59- cells in Ly6Chigh cells was higher than that in the control group on 1 d after myocardial infarction, while the proportion of P2X7+CD59+ macrophages was lower than that in the control group (all P<0.05). The percentages of P2X7+CD59+ cells in Ly6Clow macrophages at 1, 3, and 5 d after myocardial infarction were lower than those in the control group, while the proportion of P2X7-CD59+ cells was higher than that in the control group (all P<0.05). Conclusion At different stages of acute myocardial infarction in mice, the subpopulations of cardiac tissue macrophages show dynamic changes, accompanied by dynamic changes in the expressions of CD59 and P2X7.
