  • 目的  探讨外周血环状RNA hsa_circRNA_000581表达水平与急性缺血性脑卒中(AIS)痰证的关系。方法  选取2016年1月至2020年12月广西中医药大学第一附属医院收治的AIS患者171例为AIS组。选取同期与AIS组来源医院同城区的社区卫生服务中心招募的中国汉族一般人群182例为对照组。通过实时荧光定量聚合酶链反应技术检测研究对象外周血hsa_circRNA_000581相对表达水平。采用受试者工作特征曲线分析hsa_circRNA_000581表达水平对AIS的诊断价值。分析hsa_circRNA_000581在对照组和不同中医证候要素患者外周血中的表达水平,并比较AIS痰证患者hsa_circRNA_000581高表达组与低表达组的血常规及凝血功能指标。结果  AIS组外周血hsa_circRNA_000581表达水平显著低于对照组[4.43×10-4(2.90×10-4,7.14×10-4)比6.84×10-4(4.08×10-4,1.01×10-3)](P<0.001)。外周血hsa_circRNA_000581表达水平诊断AIS的曲线下面积为0.657,敏感度为69.1%、特异度为62.0%。AIS痰证组(n=68)的外周血hsa_circRNA_000581表达水平显著低于对照组(P=0.004),但AIS风证组(n=55)的hsa_circRNA_000581表达水平与对照组比较差异无统计学意义(P=0.068)。hsa_circRNA_000581低表达组(n=34)AIS痰证患者的血小板计数和纤维蛋白原水平显著高于高表达组患者(n=34),差异均有统计学意义(P=0.041、P=0.022)。结论  环状RNA hsa_circRNA_000581可能通过影响凝血功能而影响AIS痰证的发生。

  • Objective  To investigate the correlation between peripheral blood expression of circular RNA hsa_circRNA_000581 and phlegm syndrome of acute ischemic stroke (AIS). Methods  A total of 171 AIS patients admitted to the First Affiliated Hospital of Guangxi University of Chinese Medicine from January 2016 to December 2020 were selected as the AIS group. A total of 182 Chinese Han normal people recruited from community health service centers in the same urban area as the source hospital of AIS group were selected as the control group. The relative expression level of hsa_circRNA_000581 in peripheral blood was measured by real-time fluorescence quantitative polymerase chain reaction. The diagnostic value of hsa_circRNA_000581 expression level for AIS was analyzed by receiver operating characteristic curve. The hsa_circRNA_000581 expression in the peripheral blood of the control group and different syndrome elements of traditional Chinese medicine were analyzed, and the blood routine and coagulation indexes of the high and low hsa_circRNA_000581 expression groups in patients with phlegm syndrome of AIS were compared. Results  The expression level of hsa_circRNA_000581 in peripheral blood of the AIS group was significantly lower than that of the control group[4.43×10-4(2.90×10-4,7.14×10-4) vs 6.84×10-4(4.08×10-4,1.01×10-3)] (P<0.001). The area under the curve of peripheral blood hsa_circRNA_000581 expression level for diagnosing AIS was 0.657, with sensitivity of 69.1% and specificity of 62.0%. The expression level of hsa_circRNA_000581 in peripheral blood of AIS phlegm syndrome group (n=68) was significantly lower than that of control group (P=0.004). However, there was no statistically significant difference in the expression level of hsa_circRNA_000581 between the AIS wind syndrome group (n=55) and the control group (P=0.068). The levels of platelets count and fibrinogen in the hsa_circRNA_000581 low expression group (n=34) in AIS patients with phlegm syndrome were higher than those in the hsa_circRNA_000581 high expression group (n=34)(P=0.041, P=0.022). Conclusion  Circular RNA hsa_circRNA_000581 may affect the occurrence of AIS phlegm syndrome by affecting coagulation function.
