类风湿关节炎(RA)是一种以对称性多关节滑膜炎为特征的慢性全身性自身免疫性疾病。发病机制尚不清楚,缺乏有效的早期诊断标志物。目前的治疗方法只能减缓疾病的进展,不能停止或逆转它所造成的损害。长链非编码RNA(lncRNA)、环状RNA(circRNA)、微小RNA(miRNA)和信使RNA(mRNA)在内的调控网络在RA病程中发挥着重要作用。lncRNA/circRNA作为miRNA的“吸附海绵”,竞争性结合miRNA,阻碍miRNA对靶mRNA的调控,参与滑膜细胞增殖、凋亡、侵袭和炎症反应的调控。基于竞争性内源RNA角度研究RA的发病机制,有助于发现新的潜在的分子标志物、靶向治疗药物。
Rheumatoid arthritis (RA) is a chronic systemic autoimmune disease characterized by symmetrical polyarticular synovitis. The pathogenesis is still unclear, and there is a lack of effective early diagnostic markers. The current treatment methods can only slow down the progression of the disease and cannot stop or reverse the damage caused by it. Regulatory networks, including long non-coding RNA (lncRNA), circular RNA (circRNA), microRNA (miRNA) and messenger RNA (mRNA), play an important role in the course of RA. As the "adsorption sponge" of miRNA, lncRNA/circRNA competitively binds miRNA, blocks the regulation of miRNA on target mRNA, and participates in the regulation of synovial cell proliferation, apoptosis, invasion and inflammation. Researching the pathogenesis of rheumatoid arthritis from the perspective of competitive endogenous RNA can help discover new potential molecular markers and targeted therapeutic drugs.
