  • 目的 分析环状RNA ADAM22及其下游信号因子在顺铂耐药三阴性乳腺癌(TNBC)中的表达情况。方法 收集2020年1月至2021年1月新疆医科大学附属肿瘤医院病理科证实的乳腺癌患者乳腺标本,包括25例癌旁组织(对照组)和40例TNBC组织(TNBC组)。根据患者对化疗药物(多西他赛+顺铂)的敏感性,将40例TNBC组织分为化疗敏感组(18例)及化疗耐药组(22例)。采用实时荧光定量聚合酶链反应分别检测各组环状RNA ADAM22、微小RNA-29c-3p(miR-29c-3p)及色素框同源物2(CBX2) mRNA的表达量。采用Pearson相关性分析方法分析化疗耐药TNBC组织中miR-29c-3p、CBX2 mRNA与环状RNA ADAM22表达量的相关性。结果 TNBC组环状RNA ADAM22表达量明显高于对照组[(2.5±0.8)比(1.3±0.5)],化疗耐药组环状RNA ADAM22表达量明显高于化疗敏感组[(2.9±0.7)比(2.0±0.6)](均P<0.05)。TNBC组miR-29c-3p表达量明显低于对照组[(1.1±0.4)比(2.1±0.5)],化疗耐药组miR-29c-3p表达量明显低于化疗敏感组[(0.8±0.3)比(1.3±0.3)](均P<0.01)。TNBC组CBX2 mRNA表达量明显高于对照组[(2.9±0.9)比(1.5±0.5)],化疗耐药组CBX2 mRNA表达量明显高于化疗敏感组[(3.4±0.8)比(2.3±0.7)](均P<0.001)。在化疗耐药TNBC组织中,miR-29c-3p与环状RNA ADAM22表达量呈负相关(r=-0.558,P=0.007),CBX2 mRNA与环状RNA ADAM22表达量呈正相关(r=0.495,P=0.019)。结论 TNBC组织中环状RNA ADAM22高表达,且在化疗耐药TNBC组织中的表达量高于化疗敏感组织。在化疗耐药TNBC组织中,miR-29c-3p与环状RNA ADAM22表达量呈负相关,CBX2 mRNA与环状RNA ADAM22表达量呈正相关。

  • Objective To investigate the expressions of circular RNA (circRNA) ADAM22 and its downstream signaling factors in cisplatin-resistant triple-negative breast cancer(TNBC). Methods  Breast specimens from patients with breast cancer confirmed by Department of Pathology, Cancer Hospital Affiliated to Xinjiang Medical University were collected from January 2020 to January 2021, including 25 cases of para-cancerous tissues (control group) and 40 cases of TNBC tissues (TNBC group). According to the sensitivity of patients to chemotherapy drugs (docetaxel + cisplatin), 40 cases of TNBC tissues were divided into chemotherapy sensitive group (18 cases) and chemotherapy resistant group (22 cases). The expressions of circRNA ADAM22, microRNA-29c-3p (miR-29c-3p) and chromobox homolog 2 (CBX2) mRNA in each group were detected by real-time fluorescent quantitative polymerase chain reaction. The relationship among miR-29c-3p, CBX2 mRNA, and circRNA ADAM22 expressions was analyzed by Pearson correlation analysis. Results  The expression of circRNA ADAM22 in TNBC group was significantly higher than that in control group [(2.5±0.8) vs (1.3±0.5)] and the expression in chemotherapy resistant group was significantly higher than that in chemotherapy sensitive group [(2.9±0.7) vs (2.0±0.6)](both P<0.05). The expression of miR-29c-3p in TNBC group was significantly lower than that in control group [(1.1±0.4) vs (2.1±0.5)] and the expression in chemotherapy resistant group was significantly lower than that in chemotherapy sensitive group [(0.8±0.3) vs (1.3±0.3)](both P<0.01). The expression of CBX2 mRNA in TNBC group was significantly higher than that in control group [(2.9±0.9) vs (1.5±0.5)] and the expression in chemotherapy resistant group was significantly higher than that in chemotherapy sensitive group [(3.4±0.8) vs (2.3±0.7)](both P<0.001). In chemotherapy resistant TNBC tissues, there was negative correlation between miR-29c-3p and circRNA ADAM22 expressions (r=-0.558, P=0.007) and positive correlation between CBX2 mRNA and circRNA ADAM22 expressions (r=0.495, P=0.019). Conclusions  CircRNA ADAM22 is highly expressed in TNBC tissues, and the expression in chemotherapy resistant TNBC tissues is higher than that in chemotherapy sensitive tissues. In chemotherapy resistant TNBC tissues, there is negative correlation between miR-29c-3p and circRNA ADAM22 expressions and positive correlation between CBX2 mRNA and circRNA ADAM22 expressions.
