  • 目的 探讨CD147和血小板源性生长因子CCPDGF-CC)在三阴性乳腺癌(TNBC)细胞系MDA-MB-231中的表达及对细胞功能的影响。方法 采用流式细胞仪筛选MDA-MB-231CD147低表达和CD147高表达2个细胞亚群,采用免疫荧光染色检测2个细胞亚群中CD147蛋白和PDGF-CC蛋白表达情况。通过短发夹RNA干扰技术,将CD147高表达的细胞亚群分为空白对照组、Ctrl RNA组、sh-CD147组、sh-PDGF-CC组、sh-CD147+sh-PDGF-CC组。比较各组细胞增殖活性及细胞迁移功能。结果 CD147高表达细胞亚群中CD147PDGF-CC蛋白表达水平高于CD147低表达细胞亚群。与空白对照组相比,sh-CD147组、sh-PDGF-CC组及sh-CD147+sh-PDGF-CCCD147高表达细胞亚群的增殖活性更低,差异均有统计学意义(均P0.05),其中sh-CD147+sh-PDGF-CC组细胞增殖活性最低。sh-CD147组、sh-PDGF-CC组、sh-CD147+sh-PDGF-CC组的迁移细胞数量少于空白对照组和Ctrl RNA组[(20.7±4.5)、(13.7±2.5)、(7.3±2.1)个比(50.0±4.6)、(51.3±9.5)个],差异有统计学意义(F=45.130P0.001),且sh-CD147+sh-PDGF-CC组少于sh-CD147组和sh-PDGF-CC组,差异有统计学意义(F=12.910P=0.006)。结论 TNBC细胞系MDA-MB-231细胞中,CD147高表达细胞亚群的PDGF-CC蛋白表达水平也更高,下调CD147PDGF-CC蛋白的表达可起到协同抑制细胞增殖和细胞迁移功能的作用。

  • Objective To explore the expressions of CD147 and platelet-derived growth factor CC (PDGF-CC) in triple-negative breast cancer (TNBC) cell line MDA-MB-231 and their effects on cell function. Methods Flow cytometry was used to distinguish the subcellular lines between low-CD147 expression and high-CD147 expression from MDA-MB-231. Immunofluorescence staining was used to detected expressions of CD147 and PDGF-CC protein in the two subcellular lines. High-CD147 expression subcellular line was divided into Mock group, Ctrl RNA group, sh-CD147 group, sh-PDGF-CC group and sh-CD147+sh-PDGF-CC group according to short hairpin RNA interference technology. The cell proliferation activity and cell migration function were compared among the groups. Results The expressions of CD147 and PDGF-CC protein in high-CD147 expression subcellular line were higher than those in low-CD147 expression subcellular line. Compared with Mock group, the proliferative activity in sh-CD147 group, sh-PDGF-CC group and sh-CD147+sh-PDGF-CC group decreased (all P<0.05), in which sh-CD147+sh-PDGF-CC group was the lowest. The number of migrating cells in sh-CD147 group, sh-PDGF-CC group and sh-CD147+sh-PDGF-CC group was lower than that in Mock group and Ctrl RNA group[(20.7±4.5,13.7±2.5,7.3±2.1 vs 50.0±4.6,51.3±9.5)](F=45.130, P<0.001), and the number in sh-CD147+sh-PDGF-CC group was lower than that in sh-CD147 group and sh-PDGF-CC group (F=12.910, P=0.006). Conclusions  In TNBC cell line MDA-MB-231, the expression PDGF-CC is higher in high-CD147 expression subcellular line. Down-regulation of both CD147 and PDGF-CC can play a synergistic role in inhibiting cell proliferation and migration.
