目的 分析人第10号染色体缺失的磷酸酶及张力蛋白同源基因(PTEN)及胸苷激酶1(TK1)与子宫内膜癌患者预后的相关性。方法 选取中国人民解放军联勤保障部队第九二一医院及湖南省张家界市人民医院2010年1月至2015年1月诊治的子宫内膜癌患者90例为子宫内膜癌组;选取同期女性健康体检者88例为对照组。采用酶联免疫吸附试验法检测不同临床分期(临床Ⅰ~Ⅳ期)和不同病理分级(G1级、G2级和G3级)子宫内膜癌患者血清中PTEN及TK1表达水平,并分析PTEN及TK1表达水平与患者预后的相关性。结果 子宫内膜癌组PTEN/β-肌动蛋白(β-actin)水平明显低于对照组[(0.38±0.08)比(0.55±0.15)],TK1/β-actin水平明显高于对照组[(0.31±0.05)比(0.18±0.03)](均P<0.05)。子宫内膜癌临床Ⅱ、Ⅲ、Ⅳ期患者PTEN/β-actin水平明显低于Ⅰ期患者,TK1/β-actin水平明显高于Ⅰ期患者(均P<0.05)。子宫内膜癌病理分级G3级患者PTEN/β-actin水平明显低于G1级患者,TK1/β-actin水平明显高于G1级患者(均P<0.05)。随着PTEN表达的减弱及TK1表达的增强,患者的预后生存率明显降低(均P<0.05)。结论 血清PTEN及TK1水平与子宫内膜癌患者的临床分期及病理分级密切相关,TK1水平越高、PTEN水平越低,患者的预后越差。
Objective To analyze the correlation of gene of phosphatase tensin homolog deleted on chromosome 10 (PTEN), thymidine kinase 1 (TK1) and prognosis of endometrial cancer. Methods From January 2010 to January 2015, 90 patients with endometrial cancer admitted to the 921th Hospital of Joint Logistics Support Force of the Chinese People′s Liberation Army and Zhangjiajie City People′s Hospital, Hunan Province were selected as endometrial cancer group. Eighty-eight health female undergoing physical examination in the same period were selected as control group. Enzyme linked immunosorbent assay was used to detect the serum expressions of PTEN and TK1 in endometrial cancer patients with different clinical stages (clinical stage Ⅰ-Ⅳ) and pathological grades (G1, G2 and G3), and the correlation of PTEN, TK1 expressions and the prognosis of patients with endometrial cancer were analyzed. Results The level of PTEN/β-actin in endometrial cancer group was significantly lower than that in control group[(0.38±0.08) vs (0.55±0.15)], and TK1/β-actin level in endometrial cancer group was significantly higher than that in control group[(0.31±0.05) vs (0.18±0.03)](both P<0.05). The level of PTEN/β-actin in endometrial cancer patients with clinical stage Ⅱ, Ⅲ and Ⅳ was significantly lower than that in clinical stage Ⅰ, and the level of TK1/β-actin in endometrial cancer patients with clinical stage Ⅱ, Ⅲ and Ⅳ was significantly higher than that in stage Ⅰ(both P<0.05). The PTEN/β-actin level of endometrial cancer patients with pathological G3 was significantly lower than that of patients with G1, and the TK1/β-actin level of endometrial cancer patients with pathological G3 was significantly higher than that of patients with G1 (both P<0.05). With the decrease of PTEN expression and the enhancement of TK1 expression, the prognostic survival rate of patients significantly reduced (both P<0.05). Conclusion The serum levels of PTEN and TK1 are closely correlated to the clinical stage and prognosis of patients with endometrial cancer, with the higher TK1 level and the lower PTEN level, the worse the prognosis.
