目的 探讨醒脑静注射液预处理对七氟烷麻醉小鼠认知功能的影响及其作用机制。方法 选取54只成年雄性C57BL/6小鼠,采用随机数字表法分为空白组、模型组和醒脑静组,各18只。模型组和醒脑静组采用吸入3.0%七氟烷6 h建立麻醉小鼠模型,醒脑静组麻醉前2 h给予醒脑静注射液5 ml/kg腹腔注射预处理,模型组麻醉前2 h给予0.9%氯化钠注射液5 ml/kg腹腔注射,空白组不实施麻醉,与其他2组同时间给予0.9%氯化钠注射液5 ml/kg腹腔注射。对比3组小鼠实验第1、2、3天Morris水迷宫实验逃避潜伏期、60 s内穿越平台次数及实验第3天大脑海马组织中脑源性神经营养因子(BDNF)mRNA和一氧化氮合酶(NOS)的表达强度。结果 醒脑静组、模型组小鼠实验第1、2、3天Morris水迷宫实验逃避潜伏期长于空白组,醒脑静组短于模型组;醒脑静组小鼠60 s内穿越平台次数多于模型组,差异均有统计学意义(均P<0.05)。实验第3天,模型组小鼠大脑海马组织中的BDNF mRNA、NOS蛋白表达强度低于空白组[(0.53±0.12)比(1.07±0.23)、(0.12±0.05)比(0.34±0.08)],醒脑静组以上2项指标水平[(0.82±0.20)和(0.25±0.07)]高于模型组,差异均有统计学意义(均P<0.05)。结论 醒脑静注射液预处理可调节七氟烷麻醉小鼠大脑海马组织中的BDNF mRNA、NOS蛋白表达,对于减轻麻醉引起的认知功能损伤具有一定的作用。
Objective To explore the effect of Xingnaojing injection pretreatment on cognitive function of sevoflurane anesthetized mice and its mechanism. Methods Totally 54 adult male C57BL/6 mice were randomly divided into the blank group, the model group, and the Xingnaojing group, with 18 mice in each group. The mice in the model group and Xingnaojing group were anesthetized by inhalation of 3.0% sevoflurane for 6 h to establish anesthetized mice model. The Xingnaojing group was pretreated by intraperitoneal injection of Xingnaojing injection 5 ml/kg 2 h before anesthesia, the model group was given intraperitoneal injection of 0.9% sodium chloride injection 5 ml/kg 2 h before anesthesia, and the blank group was not anesthetized, which was the same time as the other two groups given intraperitoneal injection of 0.9% sodium chloride injection 5 ml/kg. The escape latency of Morris water maze test, the time of crossing platform in 60 s on the 1st, 2nd and 3rd d of the experiment and the expressions of brain derived neurotrophic factor(BDNF) mRNA and nitric oxide synthase(NOS) in the hippocampus on the 3rd d of the experiment of the three groups were compared. Results The escape latency of Morris water maze test in Xingnaojing group and model group on the 1st, 2nd and 3rd d of experiment were longer than those in blank group, while those in Xingnaojing group were shorter than those in model group; the time of crossing platform in 60 s in Xingnaojing group was more than that in model group, and the differences were statistically significant(all P<0.05). On the 3rd d of the experiment, the expressions of BDNF mRNA and NOS protein in the hippocampus of the model group were lower than those of the blank group[(0.53±0.12) vs (1.07±0.23), (0.12±0.05) vs (0.34±0.08)], and those of the Xingnaojing group[(0.82±0.20) and (0.25±0.07)] waere higher than those of the model group(all P<0.05). Conclusion Xingnaojing injection pretreatment can regulate the expressions of BDNF mRNA and NOS protein in hippocampus of mice with sevoflurane anesthesia, which has a certain effect on alleviating cognitive impairment caused by anesthesia.
