  • 目的 探讨白藜芦醇对卵巢癌细胞顺铂化疗的增敏作用和机制。方法 选取人卵巢癌SKOV3顺铂耐药细胞(SKOV3/DDP)分为对照组和观察组。对照组常规培养,不进行药物干预;观察组根据药物干预方式分为顺铂组[顺铂(32 μg/ml)干预24 h]和白藜芦醇+顺铂组[白藜芦醇(20 μg/ml+顺铂(32 μg/ml)干预24 h]。通过倒置显微镜、细胞计数试剂盒8、流式细胞仪和蛋白质印迹法分别检测3 SKOV3/DDP的生长状态、细胞抑制率、细胞凋亡情况以及B细胞淋巴瘤2家族蛋白及其相关X蛋白(Bax)和抑癌蛋白p21p21)的表达情况。结果 对照组SKOV3/DDP呈长梭形,细胞状态较好;顺铂组SKOV3/DDP出现较多圆球形的悬浮细胞,但仍有大部分细胞状态正常;白藜芦醇+顺铂组SKOV3/DDP几乎全部收缩变圆。白藜芦醇+顺铂组、顺铂组细胞抑制率、细胞凋亡率、Baxp21表达水平均高于对照组,且白藜芦醇+顺铂组均高于顺铂组[(55±4%比(41±4%、(41.52±3.24%比(30.21±1.21%、(6.2±0.7)比(4.1±0.4)、(3.69±0.39)比(2.56±0.61)],差异均有统计学意义(均P<0.05)。结论 白藜芦醇可增加SKOV3/DDP对顺铂化疗敏感性,促进SKOV3/DDP细胞内Baxp21的表达,并促进细胞凋亡。

  • Objective To investigate the sensitization effect and mechanism of resveratrol (Res) on cisplatin DDP chemotherapy in ovarian cancer cells. Methods The DDP-resistant human ovarian cancer cell SKOV3 (SKOV3/DDP) were divided into control group and observation group. The control group was cultured conventionally without drug intervention. The observation group was divided into DDP groupintervened by DDP32 μg/ml for 24 h and Res+DDP groupintervened by Res20 μg/ml+DDP32 μg/ml for 24 h. Inverted microscope, cell counting kit-8, flow cytometer and western blotting were used to detect the growth status, cell inhibition rate, apoptosis statusand, and expressions of B cell lumphoma 2Bcl-2 family protein and Bcl-2-associated X protein (Bax) and tumor suppressor protein 21(p21) of SKOV3/DDP in the three groups. Results SKOV3/DDP in control group were in long spindle shape with good cell state; SKOV3/DDP in DDP group showed more spherical suspension cells, but most of them in normal condition; SKOV3/DDP in Res+DDP group were almost completely contracted and rounded. The cell inhibition rate, apoptosis rate and expression levels of Bax and p21 in Res+DDP group and DDP group were higher than those in control group, and those in Res+DDP group were higher than those in DDP group[(55±4% vs 41±4%,41.52±3.24% vs 30.21±1.21%,6.2±0.7 vs 4.1±0.4,3.69±0.39 vs 2.56±0.61)](all P<0.05. Conclusion Res can increase the chemotherapy sensitivity of SKOV3/DDP to DDP, promote the expressions of Bax p21 and accelerate apoptosis.
