目的 观察三阴性乳腺癌患者癌组织中微小染色体维持蛋白7(MCM7)与乳腺癌特异性基因1(BCSG1)蛋白及mRNA表达水平在新辅助化疗前后的变化及相互关系。方法 选取2014年1月至2017年12月四川省德阳市人民医院乳腺外科收治的52例初治三阴性乳腺癌患者。观察新辅助化疗6个周期后的疗效,比较不同疗效患者化疗前后癌组织MCM7与BCSG1的蛋白及mRNA表达水平,并分析MCM7与BCSG1的蛋白及mRNA水平的相关性。结果 新辅助化疗后47例患者的肿块明显缩小,完全缓解12例,部分缓解35例,总缓解率为90.4%(47/52),稳定4例,进展1例。新辅助化疗后完全缓解+部分缓解组患者的MCM7与BCSG1蛋白及mRNA表达水平均较新辅助化疗前降低,差异均有统计学意义(均P<0.05),新辅助化疗后稳定+进展组患者的MCM7与BCSG1蛋白及mRNA表达水平与新辅助化疗前比较,差异均无统计学意义(均P>0.05)。新辅助化疗前后,患者MCM7与BCSG1的蛋白及mRNA水平均呈明显正相关(均P<0.05)。结论 MCM7与BCSG1的蛋白及mRNA表达水平变化能比较准确地反映肿瘤的化疗退缩情况,可作为判断三阴性乳腺癌新辅助化疗疗效的生物学指标。MCM7与BCSG1的蛋白和mRNA表达水平呈明显正相关,可以进一步探讨其在信号通路上有无密切关系。
Objective To observe the protein and mRNA changes of mini-chromosome maintenance protein 7(MCM7) and breast cancer specific gene 1(BCSG1) protein expression levels in triple negative breast cancer cells before and after neoadjuvant chemotherapy and their relationship. Methods From January 2014 to December 2017, 52 newly diagnosed triple negative breast cancer patients in Department of Breast Surgery, People′s Hospital of Deyang City, Sichuan Province were selected. The curative effects of neoadjuvant chemotherapy after 6 cycles were observed, and the protein and mRNA levels of MCM7 and BCSG1 were compared among patients with different curative effects before and after neoadjuvant chemotherapy. The correlation between MCM7 and BCSG1 on the protein and mRNA levels were analyzed. Results After neoadjuvant chemotherapy, the tumor size of 47 patients was significantly reduced. There were 12 cases completely relieved(CR) and 35 cases of partial remission(PR). The total remission rate was 90.4% (47/52), except 4 cases of stable disease(SD), and 1 case of progressive disease(PD). After neoadjuvant chemotherapy, the protein and mRNA expression levels of MCM7 and BCSG1 of patients in CR+PR group were significantly lower than those before neoadjuvant chemotherapy (all P<0.05). There were no significant differences in the protein and mRNA expression levels of MCM7 and BCSG1 in SD+PD group before and after neoadjuvant chemotherapy(all P>0.05). Before and after neoadjuvant chemotherapy, the protein and mRNA levels of MCM7 and BCSG1 were significantly positively correlated (all P<0.05). Conclusions The expression changes of protein and mRNA of MCM7 and BCSG1 can more accurately reflect the regression effect of chemotherapy that can be used as biological indicators to judge the effect of neoadjuvant chemotherapy in triple negative breast cancer. Expression levels of protein and mRNA of MCM7 and BCSG1 have obvious positive correlation, whether there is close relationship in the signal pathway can be explored next.
