KISS-1基因对人肺鳞状细胞癌细胞YTMLC-90增殖及侵袭能力的影响

  • 【摘要】目的    探讨肿瘤转移抑制基因KISS-1对人肺鳞状细胞癌细胞YTMLC-90增殖及侵袭能力的影响。方法    通过基因转染技术,将携带KISS-1基因的pEGFP-N2-KISS-1质粒转染入人YTMLC-90细胞,同时设不携带KISS-1基因的空质粒转染组和空白对照组作为对照,用蛋白质印迹法检测转染后YTMLC-90细胞中KISS-1蛋白的表达以确定转染是否成功;噻唑蓝(MTT)法测定3组YTMLC-90细胞增殖情况;Millicell小室法检测3组YTMLC-90细胞的侵袭能力。结果    成功将KISS-1基因转染入YTMLC-90细胞。pEGFP-N2-KISS-1质粒转染组YTMLC-90细胞的增殖活性与空质粒转染组和空白对照组差异无统计学意义 (P>0.05),但YTMLC-90细胞穿透Millicell聚碳酸酯膜的细胞数较空质粒转染组和空白对照组明显减少[(35±5)×105个比(82±6)×105个、(84±5)×105个],差异有统计学意义(P<0.05)。结论    KISS-1基因对YTMLC-90细胞的增殖能力没有影响,但对其侵袭能力有抑制作用,提示KISS-1基因能够抑制肺癌的侵袭。

    【Abstract】Objective     To investigate the effect of KISS-1 gene on proliferation and invasion of human lung squamous carcinoma cell line YTMLC-90. Methods    The pEGFP-N2-KISS-1 plasmid carried with KISS-1 gene and empty plasmid were respectively transfected into human YTMLC-90 cells; YTMLC-90 cells without special treatment were as a blank control group. Expression of KISS-1 protein was detected by western blot method to determine whether the transfection was successful. Proliferation activity of YTMLC-90 cells was tested by the MTT assay; invasion ability was tested by Millicell chamber method. Results    KISS-1 gene was successfully transfected into YTMLC-90 cells in pEGFP-N2-KISS-1 plasmid group. The proliferation activity in pEGFP-N2-KISS-1 plasmid group had no significant differences compared to that in empty plasmid group and blank control group(P>0.05). The number of cells that passed through polycarbonate membrane of Millicell chamber in pEGFP-N2-KISS-1 plasmid group was significantly less than that in empty plasmid group and blank control group[(35±5)×105 vs (82±6)×105, (84±5)×105](P<0.05). Conclusion    KISS-1 gene has no effect on proliferation but inhibits invasion of YTMLC-90 cells, suggesting it may inhibit the invasion of lung cancer.
